Biodegradation of remazol brilliant blue R by ligninolytic enzymatic complex produced by Pleurotus ostreatus BJM

Pleurotus ostreatus ("shimeji") is produced in Brazil on a commercial scale using various lignocellulosic residues. Efforts have been made to reuse the culture residue to obtain products of greater aggregate value such as enzymes or in processes of bioremediation. We evaluated the Remazol brilliant blue R (RBBR) degradation potential of extracts from solid substrate colonized by P. ostreatus and extracts from residue of the "shimeji" mushroom yield. Cloning, characterization and expression of a novel laccase gene Pclac2 from Phytophthora capsici BJM

Laccases are blue copper oxidases (E.C. 1.10.3.2) that catalyze the one-electron oxidation of phenolics, aromatic amines, and other electron-rich substrates with the concomitant reduction of O2 to H2O. A novel laccase gene pclac2 and its corresponding full-length cDNA were cloned and characterized from Phytophthora capsici for the first time. The 1683 bp full-length cDNA of pclac2 encoded a mature laccase protein containing 560 amino acids preceded by a signal peptide of 23 amino acids. Copper improves the production of laccase by the white-rot fungus Pleurotus pulmonarius in solid state fermentation BABT

Pleurotus pulmonarius (Fr) Quélet, produced laccase as the main ligninolytic enzyme when cultivated on solid-state cultures using corn cob as substrate. The addition of copper greatly increased the production of enzyme. The addition of 25.0 mM CuSO4 increased the level of laccase from 270 to 1,420 U.L-1 and the fungus showed high resistance to copper under the conditions used in this work.

Copper induction and differential expression of laccase in Aspergillus flavus BJM

Aspergillus flavus was isolated from soil and exhibited laccase activity under both constitutive and copper induced conditions. Spiking the medium with 1 mM copper sulfate resulted in an increase in the activity which reached 51.84 U/mL, a distinctive protein band was detected at 60 kDa. The extracellular enzyme was purified 81 fold using gel filtration chromatography and resulted in two different laccase fractions L1 and L2, the latter had a higher enzymatic activity which reached 79.57 U/mL and specific activity of 64.17 U/μg protein. The analysis of the spectrum of the L2 fraction showed a shoulder at 330 nm which is characteristic for T2/T3 copper centers; both copper and zinc were detected suggesting that this is an unconventional white laccase....

In the context of massive summer mortality events of the Pacific oyster Crassostrea gigas, the aim of this study was to investigate the early effects on genes, enzymes and haemocyte parameters implicated in immune defence mechanisms in C. gigas oysters exposed to a potentially hostile environment, i.e. to an herbicide alone or within a mixture. Following 2 h of exposure to the herbicide diuron at 1 mu g L-1 the repression of different genes implicated in immune defence mechanisms in the haemocytes and the inhibition of enzyme activities, such as laccase-type phenoloxidase (PO) in the plasma, were observed. The activities of laccase, Mn-peroxidase, phosphatase and dehydrogenase were studied in a Chilean Andisol (Temuco Serie) contaminated with pentachlorophenol (PCP). Experiments were conducted on the top of 0-20 cm soil, adding different concentrations of PCP (0.41, 4.1 and 41 mg kg-1). Enzymatic activities and residual concentrations of PCP were evaluated in soil incubated at 25 ± 1 ºC for 112 days. Enzyme activities of PCP-treated soil varied according to the PCP concentration added. In order to identify alternatives for soil fumigation in bioremediation process, the addition of benomyl and vegetable oil on the growth of L. crinitus and mitosporic fungi were evaluated. Benomyl can be used as an alternative to methyl bromide. Addition of vegetable oil favors the growth of L. crinitus.

Minimidochium parvum LPSC # 548, a fungus isolated from litter floating on waters of Río Santiago (Provincia de Buenos Aires, Argentina) polluted with industrial effluents and crude-oil, was grown as a shaking culture on a C-limited medium to evaluate its ability to produce extracellular laccase. The effect of anthracene, CuSO4 · 5H2O, ethanol, guaiacol, humic acids, Kraft lignin, MnSO4· H2O, Tween 20 and veratryl alcohol on its growth and extracellular laccase activity levels was also analyzed. Influence of pH on the growth, laccase activity and RBBR decolorization by tropical basidiomycetes BABT

The basidiomycete fungi Lentinus crinitus and Psilocybe castanella are being evaluated in a bioremediation process of soils contaminated with organochlorine industrial residues in the Baixada Santista, São Paulo. The aim of the present study was to determine the influence of pH on the fungal growth, in vitro decolorization of anthraquinonic dye Remazol Brilliant Blue R (RBBR) and laccase activity. The pH of the culture medium influenced the growth of L. crinitus and P. castanella, which presented less growth at pH 5.9 and pH 2.7, respectively. Laccase produced by a thermotolerant strain of Trametes trogii LK13 BJM

Thermophilic and thermotolerant micro-organisms strains have served as the natural source of industrially relevant and thermostable enzymes. Although some strains of the Trametes genus are thermotolerant, few Trametes strains were studied at the temperature above 30 °C until now. In this paper, the laccase activity and the mycelial growth rate for Trametes trogii LK13 are superior at 37 °C. Laccase production by Lepista sordida BJM

A Lepista sordida laccase has been characterized. Laccase and manganese peroxidase were detected in liquid medium with ammonium phosphate, yeast extract and ammonium molybdidate as nitrogen sources after 3 days of cultivation. Laccase optimal temperature and pH were 45ºC and 3.5, respectively.

Lignin degradation, ligninolytic enzymes activities and exopolysaccharide production by Grifola frondosa strains cultivated on oak sawdust BJM

Fourteen strains of Grifola frondosa (Dicks.) S. F. Gray, originating from different regions (Asia, Europe and North America) were tested for lignin degradation, ligninolytic enzyme activities, protein accumulation and exopolysaccharide production during 55 days of cultivation on oak sawdust. Lignin degradation varied from 2.6 to7.1 % of dry weight of the oak sawdust substrate among tested strains. The loss of dry matter in all screened fungi varied between 11.7 and 33.0%, and the amount of crude protein in the dry substrate varied between 0.94 to 2.55%. The strain, MBFBL 596, had the highest laccase activity (703.3 U/l), and the maximum peroxidase activity of 22.6 U/l was shown by the strain MBFBL 684. Remazol Brilliant Blue R (RBBR) dye was used as substrate to evaluate ligninolytic activity in 125 basidiomycetous fungi isolated from tropical ecosystems. The extracellular RBBR decolorizing activity produced when selected fungi were grown in solid media and in soil contaminated with organochlorines was also evaluated. A total of 106 fungi decolorized the RBBR during the growth in malt extract agar (MEA, 2%); 96 fungi showed a mycelia growth and decolorization activity stronger than the P. chrysosporium used as reference. Pentachlorophenol removal by Pleurotus pulmonarius in submerged cultures BABT

Pentachlorophenol (PCP) removal by Pleurotus pulmonarius grown in submerged cultures in the presence and absence of laccase inducers was studied in this work. When PCP was added to a final concentration of 25 mg·L-1 in submerged cultures actively producing laccase, 70% of the PCP was removed after 96 h. The removal of PCP was less than 20% in the cultures with low laccase activity. The results suggested that laccase played an important role in the biodegradation of PCP by P. pulmonarius and that for bioremediation purposes the fungus must be cultured under the conditions of active laccase production.

Physiological Characterization of Fungal Inoculum for Biotechnological Remediation of Soils BABT

The aim of this work was to study the bioremediating potential of Lentinus crinitus CCIBt2611 according to the physiological condition of the inoculum. Inoculum was prepared using sugarcane ground husk (C:N 90), at several physiological ages and applied in soil contaminated with pentachlorophenol. The inoculum's potential was assessed by evaluating the mycelium's vigor at soil's colonization, determination of peroxidase and phenoloxidase activities, in vitro degradation of Remazol Brilliant Blue R and in vivo degradation of pentachlorophenol.
